• Mutation details: 6. To develop a knock-in mouse that expresses the codon-optimized FlpO recombinase dependent on TRPV1 expression, the TGA stop codon was replaced with a 2A-Flp cassette. The targeting vector homology arms were generated by high fidelity Taq PCR using BAC clone RP23-30N20 and RP23-152H19 from the C57BL/6J library as template. The targeting vector was assembled with recombination sites and selection markers: Neomycin resistance gene (NeoR) flanked by self-deletion anchor (SDA) sites for positive selection and diphtheria toxin A fragment gene (DTA) for negative selection. The final version of the allele lacks the selection cassette. (J:363352, J:373994)
