Mutation details: The transgene comprises a modified 150 kb human HBB cluster-containing YAC A201F4.3. The modification involves the insertion of a loxP site between the LCR and the HBE1 gene and a second loxP site in opposite orientation downstream of the HBB gene and the subsequent Cre-mediated inversion of the HBB cluster with respect to the upstream LCR. Southern blot analysis confirmed the integrity and single copy number of YAC insertion. Embryonic expression of HBE1, HBG1 and HBG2 is significantly reduced compared to the non-inverted transgene.
