• Mutation details: The transgene insert was generated as follows: two differently modified mouse H19 ICR sequences (del-4 and del-5, each flanked by loxP5171 and loxP2272 sites) were inserted in reverse orientation into human HBB cluster-containing YAC A201F4.3 immediately downstream of the LCR and upstream of the HBB cluster. Subsequent Cre-mediated recombination created separate single del-4 and del-5 transgene alleles. The modifications involve the deletion of the 5' end of the ICR and all but the last 180 bp (del-4) or 240 (del-5) of a 478 bp sequence close to the 5' end that confers post-fertilization paternal methylation to the ICR.
