The Rps6ka1 gene (GenBank accession number: NM_009097.4, Ensembl: ENSMUSG00000003644) is located on mouse chromosome 4; 22 exons have been identified, with the ATG start codon in exon 1 and TGA stop codon in exon 22; the amino acid residue C564 is located in exon 18. To engineer the targeting vector, 5 homology arm and 3 homology arm was amplified from BAC DNA and confirmed by end sequencing. The C564G mutation (TGC to GGC) was introduced into exon 18 in 3 homology arm. In the targeting vector, the neo cassette was flanked by loxP sites. DTA was used for negative selection. The constitutive knock-in allele was obtained after cre-mediated recombination. (J:82809)
