Mutation details: Constitutive global C421S and C424S mutations knock-in in the mouse Rps6ka1 (Rsk1) gene. The Rps6ka1 gene (GenBank accession number: NM_009097.4, Ensembl: ENSMUSG00000003644) is located on mouse chromosome 4; 22 exons have been identified, with the ATG start codon in exon 1 and TGA stop codon in exon 22; the amino acid residues C421 and C424 are located in exon 15. To engineer the targeting vector, 5 homology arm and 3 homology arm were amplified from BAC DNA and confirmed by end sequencing. The C421S mutation (TGT to TCT) and C424S mutation (TGT to TCT) were introduced into exon 15 in 3 homology arm. In the targeting vector, the neo cassette was flanked by loxP sites. DTA was used for negative selection. The constitutive knock-in allele was obtained after cre-mediated recombination.
