Mutation detailsThe transgene contains the following: the Pomc gene, including sequence 8 kb downstream of the transcription start site and 13 kb upstream (minus the -6.5 to -0.8 kb region), containing pituitary-specific enhancer Rr281 but with neuron-specific enhancer Rr280 (nPE2) deleted (172 bp) and Rr279 (nPE1) still present, and the promoter, with the EGFP reporter gene and SV40 poly(A) signal sequences inserted upstream of the translation start site. The nPE1 enhancer core sequence was modified with three homeodomain binding sites (HDBSs) mutated from TCAAG, TAAT and ATTA to TCCCG, TCCT and ACCA. (J:224010)
