Mutation details: The SB14-31 T cell clone , specific to env122141 presented by MHC II Ab, was fused to the TCRaslphabeta-negative BW5147 thymoma cells to produce the 1A6 hybridoma cell line, which was then used as source for RNA. TCRalpha and TCRbeta cDNAs were cloned and inserted into the hCD2-VA expression cassette, which directs expression in T cells. Specifically for the TCRbeta chain, a cDNA-genomic DNA hybrid was used, in which intron 1 of the TCRVbeta-encoding gene (Tcrbv1s1) was preserved. For the present study, a TCR-transgenic line (EF4.1) expressing only the TCRbeta chain of the SB14-31 clone was selected, to maintain a polyclonal TCR repertoire.
