Mutation detailsThe targeting vector is designed to replace the endogenous STOP codon of the gene with a loxP-flanked STOP codon (TAGTGA), a TimeSTAMP cassette with a STOP codon, and an FRT-flanked neomycin (neo) cassette. The TimeSTAMP (Time-Specific Tagging for the Age Measurement of Proteins) cassette contains, from 5'; to 3', N-terminal Venus fragment, a cis-cleaving hepatitis C virus (HCV) NS3 protease, C-terminal Venus fragment, and two hemagglutinin (2xHA) tags. Flp-mediated recombination removed the neo cassette. (J:101977)
