Mutation detailsThe targeting vector is designed to insert a cassette containing an inverted enhanced green fluorescent protein (EGFP) sequence, with a polyadenylation signal, downstream of exon 1. Specifically, the vector includes, from 5' to 3', an FRT-flanked neomycin resistance (neo) cassette, an inverted EGFP flanked by inward facing 2272loxP and 5171loxP sites and third FRT site inserted downstream of the floxed EGFP. Flp-mediated recombination removed the FRT-flanked neo cassette. Cre-mediated recombination reversed the cassette trapping expression. (J:321839)
