Mutation detailsTransgenic Plasmodium berghei ANKA strain (PbA) MHC I-restricted (H2-K1 (H2-Kb)) TCR (PbT-I) mice were generated using the V(D)J segments of the TCRa- and b-genes of a CD8+ T cell hybridoma (B4) specific for P. berghei RPL6, a ribosomal protein expressed in various stages of the parasite life cycle. The B4 hybridoma is derived from a T cell line isolated from a blood-stage PbA-infected C57BL/6 mouse. The Tcra chain consists of V8S6, J17 and C2 gene segments and the Tcrb chain of V10 (V10S1A1), D2 and J2.2. The transgene also contains the Igh chain enhancer, the H2-K1 (H2-Kb) promoter and the polyadenylation signal sequence of the human beta-globulin gene for the Tcra segments and the Tcrb enhancer, the 2B4-derived 5' region and leader sequence and the B3-derived promoter and coding regions for the Tcrb segments.
