Mutation details: RMCE (recombinase-mediated cassette exchange) was utilized to insert a Myc-tagged human CEBPB sequence into the Gt(ROSA)26Sor locus. R26FNF3-1F1 (1F1) embryonic stem (ES) cells were transfected with plasmids pFLSLF2-Myc human CEPBP (carrying loxP pA-puromycin-PGK-Sv40 pA (in reverse orientation)-loxP-Myc-CEBPB-WPRE-BGH pA) and pCAG-Flpe (expressing FLP recombinase). The 1F1 ES cell line used to generate the mutation was created by targeting Gt(ROSA)26Sor with an FRT- and F3-flanked PGK-neomycin selection cassette by homologous recombination in (C57BL/6J x 129X1/SvJ)F1-derived EC7.1 ES cells.
