Mutation details: A Rosa-CAG targeting vector was designed to insert cassette containing (from 5' to 3') a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (CAG), loxP-site, Dre recombinase fused with a mutant form of estrogen receptor hormon-binding domaine C (ERT2), STOP cassette (with stop codons in all 3 reading frames and a triple polyA signal), second loxP-site, ZsGreen sequence (an enhanced green fluorescent protein engineered for brighter fluorescence and higher expression in mammalian cells), WPRE (woodchuck hepatitis virus posttranscriptional regulatory element)-polyA, and Frt flanked neomicyne selection cassette (CAG-loxp-DreERT2-stop-loxp-ZsGreen-polyA-Frt-Neo-Frt) into the Rosa26 locus.
