• Mutation details: The targeting construct includes a Tet-responsive element (TRE or tetO) and mouse prion protein promoter sequences (PrP or Prnp) directing expression of the P301L mutant variant of human microtubule-associated protein tau (MAPT) inserted into an intergenic site downstream of Col1A1. The construct, a "Flp-In" promoter cassette (PGK-promoter-ATG-FRT) and a pCAGGS-FlpE plasmid were electroporated into modified V6.5 embryonic stem cells that had previously been targeted with a Col1a1 "Flp-In" integration cassette (Col1a1-FRT-hygro-pA).
