• Mutation details: The molecular lesion is a deletion of 41 bp from exon 5 (903cgacaagatgat...gatgtgga944, Adamts10cDNA sequence, GenBankaccession number NM_172619.3), and insertion of an IRES-lacZ-neo cassette. RT-PCR (exons 3-5) shows interruption of exon 5 by insertion of the IRES-lacZ-neo cassette and loss of RNA downstream of the targeting site (exons 5-7). The fusion transcript arising from the targeted allele encodes only the ADAMTS10 propeptide and the N-terminal 13 residues of the catalytic domain. Thus, production of catalytically active ADAMTS10 is eliminated, although this could not be confirmed at the protein level in the absence of a suitable antibody. (J:275665)
