Mutation details: A loxP site and a rox site flanked hygromycin/zeocin resistance gene cassette were inserted into intron 1. Sequence coding for a 2A peptide, tdTomato cDNA, SV40 poly(A) signal sequences, an FRT site flanked flanked neomycin resistance gene cassette, a second loxP site, a splice acceptor, EGFP cDNA, and rabbit beta-globin poly(A) signal sequences were inserted into exon 12 immediately before the stop codon. Subsequent Dre- and Flp-mediated recombination excised the rox- and FRT-flanked antibiotic resistance cassettes. The intron 1 loxP site insertion disrupts enhancer Rr60779, leading to reduced transcription (78-57%) of the gene in the brain.
