Mutation details: The targeting vector is designed with a loxP-flanked transcriptional STOP cassette (PGK-puro-SV40 polyA), and an HA-tagged human DNA-damage inducible transcript 3 (DDIT3)-WPRE-BGH-pA cassette. This vector was inserted into the Gt(ROSA)26Sor locus via a FLP recombinase-mediated cassette exchange method. Specifically, the R26FNF3-1F1 (1F1) ES cell line used to generate the mutation was created by targeting Gt(ROSA)26Sor with a FRT- and F3-flanked PGK-neomycin selection cassette by homologous recombination in EC7.1 ES cells.
