Mutation details: An FRT flanked gene trap cassette composed of an FRT site, a splice acceptor, and a promoterless lacZ reporter followed by a polyadenylation signal, a loxP site, a promoter-driven neomycin resistance gene, a second polyadenylation signal and a second FRT site, was inserted via homologous recombination between exons 1 and 2, disrupting the gene. Additional loxP sites were inserted flanking exons 2 through 8 rendering this a conditional-ready allele. Splicing of exon 1 to the lacZ reporter containing the polyadenylation signal on the 3 end yields a severely truncated polypeptide if translated. Quantitative PCR analysis on cDNA isolated from gonadal and subcutaneous (inguinal) white adipose depots and kidney showed a ~50% reduction of mRNA levels in heterozygous mutant mice.
