Mutation details: A transgenic construct containing the mouse immunoglobulin heavy chain constant mu (Ighm) enhancer sequence, the human IGHM promoter, cDNA encoding mouse casein kinase 2, alpha polypeptide, the SV40 intron and polyadenylation signal, and the human IGHM enhancer was microinjected into the pronuclei of fertilized oocytes. Two independent transgenic lines, TG.CKA (A) and TG.CKB (B), were established. Ribonuclease protection analysis demonstrated expression of the transgene in the lymphoid organs of mice of both lines, more strongly in thymus than in spleen. The abundance of transgene-derived mRNA in thymus was less than 10% that of the endogenous Csnk2a transcript.
