Mutation details: The construct contains an mStrawberry red fluorescence protein fused to a modified Atg4b gene under control of a tet-responsive element (TRE; tetO). In this construct, Atg4b contains a dominant negative C74A mutation, which results in the expression of an enzymatically inactive protein. This mStrawberry/Atg4B cDNA fragment was co-electroporated into C57BL/6N-derived C2 embryonic stem (ES) cells along with a pCol-TGM targeting vector, containing an FRT-flanked PGK-Neo inserted into the 3' UTR of the Col1a1 locus, and a pCAGGS-FlpE expression plasmid.
