Mutation details: A p2lox targeting plasmid is designed to insert the human DUX4 ORF and 3' UTR followed by a poly A tail in front of a tetO promoter by exchanging it with the existing Cre recombinase ORF, using loxP and lox2272 sites. A non-functional neomycin resistance (neo) gene is placed downstream. This construct is inserted into a cassette exchange locus upstream of the Hprt gene using Inducible Cassette Exchange (ICE) recombination.
