Mutation detailsMice with this transgene express mouse mitochondrial trans-2-enoyl-CoA reductase (Mecr) under control of the mouse metallothiionein 1 (Mt1) promoter. The initial transcript comprises a fragment of the human ODC gene encompassing the 3' half of exon 1 through the 5' half of exon 3, including introns 1 and 2, which corresponds to most of the 5' untranslated region (5'-UTR) of the mature ODC mRNA, followed by the Mecr cDNA from the initiating ATG through the polyadenylation signal. Two transgenic founder mice, designated tg60 and tg62 by the authors, transmitted the transgene via the germline. Southern blot analysis reveals that both reported transgenic lines bear multiple tandem copies of the transgene. # in the transgene symbol indicates that this record represents an unspecified line number.

Mecr mRNA levels in cardiac muscle of tg60 and tg62 mice, measured by semiquantitative real-time PCR, are respectively ~1.5-fold and >12-fold that in wild-type heart; transcript levels do not differ significantly in muscle or liver of transgenic vs. wild-type mice. Administration of zinc sulfate. whether in drinking water or by intraperitoneal injection, does not further increase trangenic cardiac mRNA. Both enzymatic assay and immunoblot analysis using anti-human MECR antibody fail to demonstrate elevated MECR enzyme activity or protein level in either whole extracts or mitochondria of transgenic hearts; poor recognition of the mouse protein by the antibody and rapid turnover of the overexpressed protein are proposed explanations.
