Mutation details: The human AB775805 YAC was modified with the insertion into the 3'UTR of the HBB locus control region loxP5171 and loxP2272-flanked inverted copies of: 1) a CpG-rich 2.3 kb lambda bacteriophage DNA fragment with four CTCF-binding sites and fragment b (LCb) sequences from the H19 imprinting control region (ICR; Rr27), and 2) the H19 ICR lacking 37 bp of the Sox-Oct motifs (SOM) in fragment b. Lines 1 and 112 were generated. Line 1 was selected as the representative line. Cre-mediated recombination creates two transgenes: Tg(HBB,Rr27*)1dSOMKeij with the ICR lacking 37 bp in the SOM sequence, and Tg(HBB,lambda*)1LCbKeij with the LCb.
