Mutation details: This transgene contains from 5' to 3', the CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter, a loxP-flanked STOP cassette (beta-galactosidase/neomycin resistance cassette [beta-geo] with three copies of the SV40 polyA termination signal) and the 1.8 kbp human myelocytomatosis oncogene cDNA sequence (MYC; including exons 2 and 3 and polyA site). The 10.8 kbp transgene was linearized and transfected into 129S/SvEv-derived embryonic stem (ES) cells. An ES cell clone with a single-copy transgene integration was selected for injection into blastocysts. Constitutive beta-galactosidase (lacZ) expression is directed to multiple tissue types. Removal of the STOP cassette by Cre-mediated recombination results in removal of lacZ and expression of MYC.
