Mapping and Phenotype information for this QTL, its variants and associated markers

A genomic scan for different adipose depots (gonadal and retroperitoneal) was conducted on parental mice and a total of 457 (C57BL/6ByJ x 129P3/J) F2 mice bred from three types of reciprocal crosses: (B6 x 129) F1 females x (B6 x 129) F1 males; (129 x B6)F1 females x (129 x B6) F1 males; and (B6 x 129)F1 females x (129 x B6) F1 males. One hundred thirty nine markers were selected to span autosomes. The average distance between markers was 9.6 cM, with no gaps greater than 30 cM.

Traits used in the linkage analysis were the weight of the left and right retroperitoneal depot combined, the weight of the left and the right gonadal depot combined and the sum of both depots. Two sets of fat depot weight indexes were calculated: 1) absolute values unadjusted for body size and 2) relative values adjusted for body size (bs). Since adipose depot weight differ between male and female mice the data was also analyzed separately by sex in adition to the combined samples.

Three types of results were found:

1) suggestive evidence only for absolute adipose weight on Chrs 5, 11, 14, and 17;

2) suggestive linkage only for relative adipose weight on Chrs 8, 9, 15 and 16;

3) and suggestive linkages for both on Chrs 2, 7 and 12.

A gonadal adipose QTL on Chromosome 2, representing a relative measurement of the phenotype adjusted for body size, mapping to 91 cM in a confidence interval spanning between 56-110 cM has been labeled Adip26. This QTL has a LOD score of 3.5 in both male and female mice and accounts for 9.5 % of the variance. Heritablity was not sex dependant at this locus, the mode of inheritance was free and the 129P3/J allele was reported as the plus alelle.

Another QTL on Chromosome 7, also representing a relative measurement of the gonadal adipose phenotype adjusted for body size, has been labeled Adip27. This QTL maps to a peak at 32 cM in a confidence interval spanning between 0-42 cM, with a LOD score of 3.8 for both male and female mice, accounting for 10.2% of the variance. The C57BL/6ByJ allele was reported as the plus allele at this locus.
