Mutation details: The transgene contains the SMP8 promoter fragment (including 1074 base pairs of 5' flanking DNA, the 48 bp exon 1, 2.5-kb intron 1 and 15 bp exon 2) from the mouse actin, alpha 2, smooth muscle, aorta gene (Acta2) driving expression of a cDNA encoding human amine oxidase, copper containing 3 (AOC3; also called SSAO, semicarbazide-sensitive amine oxidase), which is followed by the SV40 late polyadenylation signal. Northern blot analysis demonstrates high-level expression of transgene-derived mRNA in stomach, bladder and intestine of all four transgenic mouse lines analyzed and at lower levels in muscle, spleen, heart, brain and kidney, but not in liver, of most lines. AOC3/SSAO activity in sera of 3-4 month old transgenic mice ranges from 8- to 52-fold that of nontransgenic littermates. Enzyme activity is elevated relative to control values in transgenic aorta, lung and abdominal adipose tissue and somewhat in kidney, but not in pancreas.

Ten independent germline transmitting transgenic mouse lines, designated lines 12, 22, 33, 43, 46, 56, 72, 77, 86 and 87, were generated and subjected to preliminary expression analysis. Mice of lines 12, 56, 72 and 86 were further characterized, those of lines 12 and 86 in greatest detail. ( J:135394)
