Mutation detailsThis transgene consists of a 2.4 kb fragment containing exons 2 and 3 of the murine c-myc oncogene under the control of the rat L-type pyruvate kinase gene regulatory region. The 3' UTR of the Myc gene was replaced with the 3' UTR region of SV40 early gene to enhance stability of myc mRNA. Transcription of Pklr is dependent on metabolic and hormonal controls, such that glucose and insulin increase transcription, while fasting, refeeding a low-carbohydrate diet and glucagon inhibit Pklr transcription. Thus manipulation of diet can result in expression of the transgene. Line 73 carries approximately 100 copies of the transgene. Northern blot analysis indicates transgene expression in the liver and to a lesser degree in the small intestine and kidney. (J:185488)
