Mutation detailsThe following elements were inserted, in order, 2 kb 5' of the Ubb gene by targeted recombination in 129-derived ES cells: CMV enhancer/beta-actin promoter (CAG), loxP site, beta-geo, 3 transcription sites, loxP site, mitochondria targeted GFP fused to a C-terminal V5 epitope, and a bovine growth hormone 3' UTR. Mitochondrial targeting was achieved by using the N-terminal 25 amino acid targeting signal of mouse Cox8a. The V5 epitope encodes amino acid residues 95-108 (GKPIPNPLLGLDST) of RNA polymerase alpha subunit of simian virus 5. (J:101977)
