Mutation detailsThe transgene contains the promoter of the rat enolase 2, gamma neuronal gene driving expression of a cDNA encoding the beta subunit of the rat glycine receptor, which is followed by the SV40 small T-antigen splice and polyadenylation signals. Brains of heterozygous transgenic mice of line 416 on a homozygous Glrbspa background contain transgene-derived mRNA at ~16% and total exon 4- and 5-containing mRNA at ~25% of the level of endogenous Glrb mRNA present in brains of wild-type mice, as visualized by northern blot analysis. In-situ hybridization demonstrates widespread expression in the brain, including the brain stem, and particularly in the facial nucleus and superior olivary complex. Western blot analysis demonstrates presence of membrane-bound glycine receptor subunit alpha, a proxy for the beta subunit. (J:32212)

Four transgenic founder lines were derived. Mice of line 416. which exhibited the highest expression, demonstrated complete rescue of the neurologic and reproductive phenotypes of Glrbspa (J:32212).
