Mutation details: Mice bearing this transgene express the normal, cellular mouse prion protein under control of the promoter of the enolase 2, gamma neuronal gene (Eno2). Of the prion protein gene (Prnp), it contains the first exon, first intron, and second exon followed immediately by the third exon, which contains the entire Prnp open reading frame (ORF), and the 3' untranslated region. This construct was inserted between the first intron and the second exon of the enolase 2 gene in a construct containing 2.8 kb of 5' flanking DNA, exon1, intron 1, and exon 2 (followed by the SV40 polyadenylation signal). Mice hemizygous for this transgene on a background lacking a functional endogenous prion protein gene express the transgene in neurons, including cerebellar granule cells (CGC), at higher levels than observed for the endogenous gene in wild-type mice.
