Mutation details: The transgene comprises one to two copies of bacterial artificial chromosome (BAC) clone RP23-291P1, whose 182-kb, C57BL/6J-derived genomic DNA insert contains the Abc11 gene, 33 kb of 5' flanking and 44 kb of 3' flanking DNA, and the entire glucose-6-phosphatase, catalytic, 2 (G6pc2) gene. Northern blot analysis of total liver RNA and western blot analysis of canalicular liver plasma membrane extracts showed about two-fold elevation of Abc11 transcript and protein levels in transgenic mice fed a chow diet; transcript levels were unaffected by consumption of a lithogenic diet for 8 weeks.
