Mutation detailsMice with this transgene inducibly express human nuclear receptor subfamily 3 group C member 1 (NR3C1; also called glucocorticoid receptor alpha) and E. coli beta-galactosidase from a bidirectional tetracycline operator- (tetO-) driven promoter. The transgene construct was generated by insertion of the full length NR3C1 coding sequence, followed by the bovine growth hormone polyadenylation signal, into the multiple cloning site (MCS) of a modified pBI-3 vector. This vector contains the bidirectional Pbi-1 promoter, consisting of the tetracycline responsive element, comprising 7 copies of the tetracycline operator sequence (tetO) separated by spacer DNA sequences, flanked by two oppositely-oriented human cytomegalovirus (hCMV) minimal promoter sequences. One promoter (Pmin-2) drives expression of the E. coli lacZ gene fused at its 5' end to a sequence encoding the SV40 nuclear localization signal and followed by the rabbit beta-globin intron and polyadenylation signal. The second promoter (Pmin-1) is followed by a multiple cloning site into which the NR3C1 coding sequence has been inserted.
