Mutation details: The first intron of the endogenous gene is interrupted by the U3Ceo secretory gene trap vector. This vector comprises an enhancerless, promoterless Moloney mouse leukemia (retro)virus genome whose LTR U3 region contains the Ceo fusion gene, which encodes a protein joining the promoterless type II transmembrane and extracellular domains of human CD2 to bacterial neomycin phosphotransferase; the CD2-encoding DNA segment includes a cryptic splice acceptor site. Expression of the endogenous gene is undetectable by northern blot analysis of total brain RNA from homozygous mutant mice.
