A series of linked genetic loci on mouse Chromosome 17 associated with obesity traits were identified by analysis of seven subcongenic lines derived from C57BL/6J-Chr 17A/J/NaJ. This consomic line carries an A/J-derived chromosome 17 on a C57BL/6J genetic background and is resistant to obesity, steatosis (fatty liver) and insulin resistance on a high-fat simple carbohydrate (HFSC) diet whereas background strain C57BL/6J is susceptible. Seven different subcongenic lines were created by backcrossing C57BL/6J-Chr 17A/J/NaJ to C57BL/6J and selecting for recombinants. Male subcongenic animals were placed on HFSC diet or low-fat simple carbohydrate (LFSC) diet at 5 weeks of age for a duration of 16 weeks. Body weight and food intake were measured weekly until time of sacrifice when plasma lipids were also analyzed.

Obrq13 (obesity resistance QTL 13) mapped to a 28.7 Mb interval between D17Mit113 (6.5 cM) and D17Mit133 (10.4 cM). A/J-derived alleles at Obrq13 confer resistance to diet-induced obesity, steatosis, decreased plasma cholesterol, decreased fasting plasma glucose, decreased fasting plasma insulin and decreased insulin resistance on a HFSC diet. On the LFSC diet, AJ-derived alleles at Obrq13 confer decreased plasma triglyceride levels and resistance to steatosis. Potential candidate genes in the Obrq13 interval include Igf2r (7.35 cM), Igfals (17.1 cM), Acat2 (7.55 cM), Acat3 (7.55 cM), Tulp4 (3.4 cM) and Lmf1 (11.5 cM). Previously identified obesity QTL mapping near Obrq13 include Adip18 (10 cM), Obq4 (4 cM) and Obq19 (8 cM).

Obrq14 (obesity resistance QTL 14) mapped to a narrow interval on chromosome 17 between 24.5 cM (D17Mit178) and 30.2 cM (D17Mit139). A/J-derived alleles at Obrq14 confer increased body weight, steatosis susceptibility, increased plasma cholesterol and increased insulin resistance on the HFSC diet. Potential candidate genes located within the Obrq14 interval include Ndufa11, Mrpl14, Mrpl2, Mrps10 and Mrps18. Obwq4 (32 cM) is a previously identified obesity QTL mapping nearObrq14.

Obrq15 (obesity resistance QTL 15) localized to a 32 Mb interval between D17Mit20 (34.3 cM) and D17Mit2 (49.7 cM). A/J-derived alleles at Obrq15 confer increased levels of plasma free fatty acids and resistance to obesity on the HFSC diet. Potential candidate genes located within the Obrq15 interval include Lpin2, Ppm1b (50.8 cM) and Tgif1.

Obrq16 (obesity resistance QTL 16) was identified at 55.7 cM near D17Mit155. A/J-derived alleles at Obrq16 confer decreased hepatic triglyceride levels on the LFSCdiet.

Obrq17 (obesity resistance QTL 17) is located at 16 cM on chromosome 17 near D17Mit198. A/J-derived alleles at Obrq17 confer decreased plasma triglyceride levels on the LFSC diet.
