Mutation details: The transgene construct contains a 650-bp segment of the promoter region of the rat insulin II gene that is truncated 30 bp upstream of the initiation codon on exon 1, which drives expression of a 568-bp EcoRI-StyI
cDNA fragment of the human PTHLH(1-141) cDNA. This sequence
contains the entire pre-pro and coding region sequences of the
PTHLH(1-141) isoform but only 20 bp of 39 untranslated sequence. At the 3' end of the transgene construct, human growth hormone sequences containing transcription termination, polyadenylation, and splicing signals were added.
RNAase protection assays and immunohistochemical staining confirmed the pancreatic overexpression of the human mRNA and protein, respectively.
