Mutation detailsA targeting vector was designed to substitute alanine for serine at positions 927 and 933 (S927A, S932A). Targeting took place in PC3 ES cells in which Cre recombinase expression is restricted to haploid spermatids. A loxP-flanked neomycin resistance cassette was included upstream of exon 24 for targeting purposes and was subsequently removed by Cre mediated recombination upon transmission through male chimeric mice. This mutation was predicted to prevent phosphorylation of the gene product by IKK making it resistant to signal-induced degradation. (J:142340)
