Mutation detailsAlternative splicing of this locus produces different protein products. One such protein called small MBL-associated protein (sMAP) has its C-terminal end encoded by exon 5. This exon is not believed to be used by the other alternative transcipts of this gene locus. A floxed neomycin-resistance cassette replaced exon 5 of the loci and an additional loxP site was inserted into the promoter region for future conditional targeting of the first 4 exons. Homozygous mice do not produce the sMAP transcript as determined by Northern blot and quantitative RT-PCR analysis on liver extract. However, two other alternative transcripts associated with this locus, H and L, had their expression reduced to 2% of wild-type. Immunoblotting of serum failed to detect any of the transcripts.
