Mutation details: A targeting vector was designed to introduce a loxP-flanked neomycin-resistance cassette in reverse transcriptional orientation to intron 17 and an additional loxP site to intron 18. Exon 18 encodes the transmembrane domain of the gene (also called CD148). The targeting construct was transfected into E14.1TG3B1 129P2/OlaHsd-derived embryonic stem (ES) cells. Targeted animals were bred with BALB/c-background CMV-Cre transgenic female mice. Progeny lacking the neomycin cassette, but retaining exon 18 flanked by loxP sites were bred to remove the CMV-Cre transgene.
