• Mutation details: A targeting vector was constructed where lysines 213 and 214 that are part of the ATP-binding pocket of the protein were mutated to methionines (KK213-214MM). An FRT-flanked neomycin selection cassette was also included and then subsequently removed leaving behind a single FRT site. Western blots confirmed the mutant protein has similar expression to wild-type protein. Abolition of kinase activity of the protein was confirmed by an in vitro assay. (J:125732)
