Mutation details: The construct consisted of the sequence expressing human alpha-synuclein 1-120 which is truncated, lacking the final 20 amino acids inserted downstream of the mouse prion protein promoter. These mice were crossed to C57BL/6JOlaHsd mice which lack alpha-synuclein as a result of a spontaneous deletion of the alpha-SNCA locus. Transgenic expression of the truncated protein is more widespread in the substantia nigra and striatum than that observed in mice with the same transgene expressed by the tyrosine hydroxylase promoter.
