Mutation detailsTo generate the I-Ab-restricted CD4+ 1H3.1 T cell hybridoma, a peptide corresponding to residues 52-68 of the a-chain of the I-E MHC class II molecule (Ea52-68) was injected into C57BL/6 mice. T cells expressing the T cell receptor (TCR) combination (Va1-Ja21 and Vb6-Db2.1-Jb2.6) were selected. These TCR sequences were cloned into TCR expression cassettes and the resulting constructs were co-microinjected into (C57BL/6 x SJL)F2 oocytes. Mature T cells from 1H3.1 TCR Tg mice (Va1Vb6) are skewed toward expression of the CD4 coreceptor. A subset of TCRhigh CD8+ T cells (5-10%) is found in the periphery. (J:124574)
