Mutation detailsA neo cassette replaced the entire gene as well as 3.3 kb sequence upstream and 3.5 kb sequence downstream of the gene. RT-PCR analysis determined that expression of adjacent genes was not affected. Analysis of 129 derived sperm from chimeras showed reduced mRNA and protein expression, as determined by quantitative PCR and western analysis. (J:100434)

Authors state that haploinsufficiency of gene prevents genetic transmission of the mutated allele and generation of non-chimeric heterozygous mice. This phenotype differs from Cyp17a1tm1Mell heterozygous male mice, which are fertile and have germline transmission of the mutant allele.
