Mutation detailsThe construct contains a 1.35 kb porcine transforming growth factor beta 1 (TGFB1) cDNA inserted into the first exon of the mouse glial fibrillary acidic protein (Gfap) gene, making a fusion gene The TGFB1 cDNA has been mutated to encode serines at positions 223 and 225, instead of cysteines, to ensure secretion of a bioactive peptide. The linear construct was microinjected into (BALB/c x SJL)F2 one-cell stage embryos. Line 64 founder mice expressed low levels of TGFB1 mRNA in the brain. Line 115 was also generated and shows very similar properties. (J:27781)

Notes
Homozygous trangenic mice develop severe hydrocephalus within 7-20 days after birth and exhibit dwarfism relative to non-transgenic littermates. Mice develop seizures, tremors, incoordination, and hunched postures. Only 1% of heterozygotes developed hydrocephalus.
