Mutation detailsThe transgene comprises a 15.5 kb DNA fragment of clone 1B-1 from a lambda phage library of liver genomic DNA from the M. m. molossinus-derived inbred strain MSM that includes a 5,420 bp sequence containing all the coding exons and introns, 3.8 kb of 5' and 6.3 kb of 3' flanking DNA of a non-classical MHC Class I gene of the H2-T group. Although expression of the native gene in MSM mice is restricted to thymus and intestine, northern blot analysis detects transcripts in thymus, intestine, spleen, lymph nodes, bone marrow and testis of transgenic mice. FACS analysis with anti-TL antibody demonstrated expression of the protein in cells from thymus, lymph nodes and spleen of transgenic mice. (J:19193, J:113017)

Notes
This is one of three transgenic lines made in the C3H/He background, which lacks thymocyte expression of H2-T antigen, using this construct. Line 5 has approximately 8, line 51 approximately 20, and line 8 approximately 2 copies of the transgenic construct per haploid genome, The three transgenic strains exhibit the same tissue distribution of mRNA and protein expression at levels roughly proportional to their transgene copy numbers.
