Mutation details: The transgene expresses the reverse tetracycline regulated transactivator gene, rtTA, under direction of a control sequence derived from the promoter and upstream regulatory elements of the rat fatty acid binding protein 1 gene. The control sequence consists of nucleotides (nt) -596 through +21 of the rat gene, with 4 additional copies of nt -177 through -133 inserted following nt -132. Following the control sequence is the human growth hormone gene, beginning at nt +1 and interrupted immediately after the translation initiation codon by the rtTA coding sequence, which is followed by a series of stop codons. RNase protection analysis demonstrates rtTA expression specifically in epithelium of the small intestine, cecum and colon; transcript levels are very low in the duodenum, increase through the jejunum, ileum and cecum, and are low in the colon. No mRNA is detected in bladder or other extraintestinal tissues analyzed.
