Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 263 (A/J x C57BL/6J)F1 x A/J backcross animals to identify loci associated with resistance to malaria. 145 polymorphic markers at an average spacing of 5 cM - 15 cM were used in the genome scan. Animals were infected with Plasmodium chabaudi and assessed daily for parasitemia. Parental strain A/J is susceptible to P. chabaudi whereas parental strain C57BL/6J is resistant. F1 hybrids are resistant similar to C57BL/6J.

Linkage to malaria resistance mapped to mouse Chromosome 8 and is named Char2 (P. chabaudi malaria resistance QTL 2).

mapping population in 1997 in J:44414, which differs from the population used here, we consider this a separate mapping experiment and have named the QTL Char11, P. chabaudi malaria resistance QTL 11.

C57BL/6J-derived alleles at Char11 confer resistance to malaria with a dominant mode of inheritance. Char11 was confirmed in 143 (A/J x C57BL/6J)F2 intercross animals. Genetic markers associated with parasitemia are D8Mit25 (32 cM), D8Mit195 (37.5 cM), and D8Mit41 (41 cM), with peak linkage occurring at D8Mit25 (LOD=4.3). Char11 accounts for 10% of the phenotypic variation. Potential candidate genes for Char11 include Gypa (36 cM), Ea1 (38 cM),Erp1 (43 cM), Il15 (38 cM), and Msr1 (20 cM, formerly Scvr).

Suggestive linkage to parasitemia was also detected at D5Mit140 (70 cM), D15Mit11 (10.4 cM), and D17Mit16 (17.4 cM) in the backcross but were not confirmed in the F2 population.
