Notes

This transgene was designed to permit assessment of mutation frequencies in the host mouse. The pUR288 plasmid copies are excised from mouse genomic DNA by restriction endonuclease digestion, recircularized and introduced into E. coli C lacZ^- cells. Selection by ampicillin, X-gal and P-gal identifies colonies bearing pUR288 containing mutant lacZ genes. The mutant plasmids can then be isolated for characterization using lacI repressor protein-coupled magnetic beads.
