Mutation details: Exons 2 and 3 were replaced with the En2A-IRESbetageoPA cassette. The replaced sequence encodes amino acids 194 to 452, encompassing the entire catalytic domain of the protein, including the protein tyrosine phosphatase active-site motif HCXXGXXRS. Western blot failed to detect protein in mutant ES cells.
