Mutation detailsThis transgenic line carries 8-10 copies of a construct comprising 254 base pairs of 5' flanking DNA and 106 base pairs of untranslated sequence from exon I of the human hexokinase II gene joined to the firefly luciferase coding sequence, which is followed by the SV40 small intron and polyadenylation signal. No distinction was made among the 5 transgenic lines - this and Tg(HK2-luc)229Uku, 230Uku, 232Uku and 234Uku - carrying from 1-2 to 45-50 copies of the same construct; the authors state there were no significant differences among them, whether or not results were normalized to copy number. It was noted that baseline luciferase expression varied inversely with transgene copy number. Baseline luciferase activity in heart, skeletal muscle and adipose tissue of 4 to 5-week old mice bearing these transgenes was not statistically greater than background.
