Mutation details: Exon 1 was replaced with cDNA encoding a human endothelial receptor type B fused to the C-terminal 10 amino acids of rhodopsin immediately before the stop codon and 0.9 kb of the 3' UTR sequence with three pA signals and a PGK-neo selection cassette. Northern blot of mutant demonstrated successful expression of the knock-in sequence under endogenous control sequences. Transcripts of three different lengths were detected, corresponding to location of the three pA signals. Quantitative RT-PCR detected levels of mutant transcripts were present at ~8.7% of wild-type levels.
