Mutation detailsThe transgene comprises a Sfpi1 minigene under transcriptional control of the long terminal repeat (LTR) of the spleen focus-forming virus (SFFV) responsible for Friend erythroleukemia. The minigene was composed of genomic DNA fragments containing exons 1 and 2 and the 5' end of exon 3, each exon flanked by some contiguous DNA (100 - 500 base pairs) from its neighboring intron(s), joined to a partial cDNA derived from the remainder of exon 3 plus exons 4 and 5. This minigene, which includes the endogeous translation initiation and termination codons and polyadenylation signal, was ligated to a fragment of the SFFV LTR that included U3, R and 20 base pairs of U5.
